SDS Sample Buffer

(also known as Laemmli Buffer or Tris-Glycine SDS Sample Loading Buffer)

1X SDS Sample Buffer consist of:
63 mM Tris HCl
10% Glycerol
2% SDS
0.0025% Bromophenol Blue
pH 6.8

Reference for Laemmli Buffer:
Laemmli, Nature 227:680-5, 1970.

The following recipe is for 2X SDS Sample Buffer:

0.5 M Tris-HCl, pH 6.8 2.5 ml
Glycerol 2 ml
10% (w/v) SDS 4 ml
0.1% (w/v) Bromophenol Blue    0.5 ml

Bring the volume to 10 ml with ultra pure water.

Add Beta-Mercaptoethanol (2-Mercaptoethanol) at a final concentration of 2.5 to 5.0%. Beta-Mercaptoethanol is necessary to reduce intra or intermolecular disulfide bonds.
Denature samples by heating at 85 C for 2-5 min.
