Technical Specifications

Calcium flux assays are preferred methods in drug discovery for the detection of G-protein-coupled receptors (GPCRs). The Screen Quest Fluo-8 NW Calcium Assay Kit provides a homogeneous fluorescence-based assay to detect intracellular calcium mobilization. Cells expressing a GPCR of interest that signals through calcium are preloaded with our proprietary Fluo-8 NW that can penetrate the cell membrane. Fluo-8 NW is the brightest calcium indicator available for the detection of HTS.

Once inside the cell, the lipophilic blocking groups of Fluo-8 AM are cleaved by nonspecific cellular esterase, resulting in a negatively charged fluorescent dye that remains inside the cells, and its fluorescence is greatly increased by binding to calcium. . When cells are stimulated with detection compounds, the receptor signals intracellular calcium release, greatly increasing Fluo-8 NW fluorescence.

The characteristics of its long wavelength, high sensitivity, and fluorescence increases of 100 to 250 times (when complexed with calcium) make Fluo-8 NW an ideal indicator for the measurement of cellular calcium. This Screen Quest Fluo-8 NW Calcium Assay Kit provides an optimized assay method for monitoring G-protein-coupled receptors (GPCRs) and calcium channels. The assay can be performed in a convenient 96- or 384-well microtiter plate format and can be easily adapted for automation.
