Description:

The monoclonal antibody SER-4 reacts with mouse CD169, also known as Sialoadhesin and Siglec-1. CD169 is a 185 kDa member of the Siglec family of endocytic and adhesion receptors that recognize sialylated glycans. This receptor is expressed on a subset of macrophages that are found with high frequency in the spleen, liver, lymph nodes, and bone marrow.

CD169 can also be detected on monocytes in the bone marrow and periphery. This receptor plays a role in cell-cell adhesion & nbsp; as well as in the internalization of sialylated viruses and bacteria. This endocytic pathway allows CD169 + macrophages to present lipid antigens to iNKT cells, leading to their activation. Studies have also shown that the direct interaction of CD169 with regulatory T cells inhibits their proliferation. The SER-4 antibody has been reported to have antagonistic activity.

Informed Applications:

This SER-4 antibody has been reported to be used in flow cytometric analysis.

Proven Applications:

This SER-4 antibody has been tested by flow cytometric analysis of mouse bone marrow cells. This can be used at a dose less than or equal to 0.25 µg per test. A test is defined as the amount (µg) of antibody that stains a cell sample in a final volume of 100 µL. The number of cells must be determined empirically, but can range from 10 ^ 5 to 10 ^ 8 cells/test. It is recommended that the antibody be carefully titrated for optimal performance in the assay of interest.

eflour 660 is a replacement for Alexa Fluor 647. eflour 660 emits at 659 nm and is excited by the red laser (633 nm). Make sure your instrument is capable of detecting this fluoroscopic.

Destination information

Siglecs are a family of membrane-bound lectins (from the immunoglobulin superfamily) that bind to sialic acid and mediate cell-cell interactions. Members of the family include sialoadhesin, CD22, and CD33. Sialoadhesin is a macrophage-specific cell adhesion molecule that is expressed on activated macrophages in chronic inflammation and tumors.
